Testing Procedure Of Manganese Testing in Water Sample in our lab

Manganese testing in water systems is crucial for water quality management. Manganese is an essential nutrient but it is consuming excessive amounts is highly toxic.Manganese can effect the flavour and colour of water. 

Here we take Drinking Water to test the manganese,followed Periodate colorimetric Method.

  • Dilute Sulphuric Acid(1:1) :Provides the acidic medium necessary for oxidation, prevent precipitation of manganese compounds.To prepare a (1:1) dilute sulfuric acid solution, mix equal parts by volume of concentrated Sulphuric Acid and distilled water.
  • Nitric Acid : Nitric Acid is used in drinking water testing to digest and break down organic matter.
  • Distilled Water: Prevents contamination from trace metals or ions that may interfere with color development.The conductivity of distilled water should be <5 μS/cm to prevent contamination.
  • Phosphoric Acid: Phosphoric acid provides the necessary acidic environment to prevent side reactions.
  • Potassium Periodate: In drinking water testing, Potassium Periodate is used as a powerful oxidizing agent in the standard periodate colorimetric method.
  • Standard Manganese Solution: Solutions of known manganese concentration are measured to establish the relationship between absorbance and concentration.Dissolve 3.077 g of manganese sulfate monohydrate in distilled water and diluting the solution to 1 liter in a volumetric flask. A few drops of dilute sulfuric acid may be added for stability.
  • Volumetric Flask(1litre,50 ml): Used for precise dilution.
  • Pipettes(1ml & 2ml)-Used for transferring exact volumes of reagents and samples.
  • UV-Vis Spectrophotometer :Measures the absorbance at a wavelength of around 525 nm.
  • Hot Plate :For boiling the sample after acidification to dissolve soluble matter.

To a 500 ml conical flask measure a suitable volume(250 ml) of the well mixed sample .


Add 2-3 ml mixture of nitric acid & 1 ml dilute sulphuric acid and heat the mixture,until all traces of organic matter are completely destroyed.


After that, Add 10 ml distilled Water, 5 ml of the phosphoric acid and 0.2 gm potassium periodate ,bring to the boil until it reduces to 30 ml.


Cool the mixture in room temperature.


Transfer into a volumetric flask ,adjust the volume to 50 ml with distilled water and mix well.


If manganese is present ,it turns into pink colour to violet.


Take the absorbance at 525nm through spectrophotometer.

Calculation:

Manganese(mg/L) = (Sample Absorbance-Blank Absorbance) x Calibration Factor/ Weight of Sample
